Science and Beers: Ultraminiaturization in Drug Discovery

Plants, bacteria, and animals synthesize an incredible array of diverse bioactive compounds orchestrated through tight spatial and temporal metabolic control.

Compartmentalization of chemical reactions ensures optimal conditions for the regulation of enzymatic cascade (and multi-step) reactions in cellular environments and allows efficient recycling of catalysts (enzymes) and individually reaction conditions for each compartment. It has long been a dream of chemists and synthetic biologist to mimic, understand and monitor such multi-step processes on the single compartment level, which corresponds to microlitres, nanolitres and attoliter volumes as the ultimate frontier for the smallest addressable (barcoded) volumes for chemistry known

The biggest obstacle to all current studies has been the lack of easily addressable biomimetic compartments (e.g., liposomes = lipid nanoreactors) and the technology for encoded leakage free fusion of multiple lipid nano-compartments in sequential or parallel fashion. The presented technology will use DNA-programmed and encoded "fusion" of lipid nano-compartments for multistep cascade reactions.

The presented technology combines frontier chemistry in aqueous media (water) for green chemistry on an ultraminiaturized (atto- or zeptoliter volumes) nanoreactor platform for combinatorial chemistry, including classical synthetic and enzymatic cascade reactions applied to accelerated discovery of new drug candidates

The technology platform is developed towards accelerated discovery of new organic compounds
for applications in drug discovery, food science, chemical biology, and pharmaceutical sciences.

Stefan Vogel is a medicinal chemist, Ph.D., and Associate Professor at the Department of Physics, Chemistry, and Pharmacy at the University of Southern Denmark. He recently received a grant from the Novo Nordisk Foundation for DKK 14 million for the NERD project for developing and applying a new platform technology for DNA-controlled molecular libraries in nanoreactors
